１と２は、単数形の instruction 、 ３は、複数形のinstructions です。
と言う事は、単数と複数で意味が異なるという事です。
では、まず、単数形の１と２を見ると：
１．（数えられる名詞） An instruction （ひとつのインストラクション）は、誰かが人にするように言うことである。
例：二人の弁護士は、（外に出るという）指示がなければ、その建物から出ないように言われた。
２．（数えられない名詞） 誰かが、ある科目や技術についてインストラクションを与えるとすれば、その科目や技術を教えることだ。
例：各候補者は、安全教育を受けている。
３． (複数形） 明瞭で細部に渡った何かの仕方についての情報である。
例：薬を飲み始める前に、必ず説明書を読みなさい。
