Do I live to smile or I smile to live?

(b^-゜) Okay people, if anyone has bothered to open the file for the e-practical, you would have noticed that the teacher has ever so intelligently created a sound recording for the procedure part without subtitles. To prevent such an ordeal to happen to every single one of us having to listen to his damned sexy voice over and over again, I have already taken down word for word of what he has said in the flash animation.


So people, here are the procedures, raw with totally no edited grammar on my part:


1. Wearing gloves, dice the medium size onion into cubes no larger than 3mm. The gloves prevent DNA enzymes on your hands from cutting the DNA into the small fragments so that it will not spool.


2. Weigh out 2.0g of diced onion, transfer all of the weighed material to a 250mL beaker.


3. Add 30mL of homogenising medium to the diced onion and incubate the beaker in 60C waterbath for 15 mins.


4. Quickly cool your preperation to 15-20C in an ice bath. This step should be accomplished in about 6 mins and prevents the denaturation of DNA.


5. Homogenise for 45 secs at low speed, followed by 30 secs at high speed.


6. Transfer 1.5mL of the homogenised mix to a 2mL tube and cetrifuge at 500 rounds/min for 5 minutes to seperate the cell debris from the homogenate.


7. Transfer the homogenate into a 10mL test tube.


8. Measure out 8mL of ice cold ethanol into a cold graduated cylinder, slowly add the ethanol down the side of the test tube until the white stringy DNA precipitate appears.


9. Spool out or wind out the stringy DNA onto a glass rod by rotating the rod in one direction only in the beaker of DNA; Continue to rotate the rod as you move it in large circles through the beaker.


10. If the DNA is intact, you should have a fluffy stringy mask spooled on one end of the glass rod.
